Target Information
Mertansine (DM1, emtansine)은 티올기를 포함한 메이탄시노이드(maytansinoid)로, 항체-약물 접합체(ADC)를 형성하기 위해 단클론 항체에 결합됩니다. DM1은 미세소관 형성을 억제하는 튜불린 저해제로, 종양 세포 내에서 표적 단백질에 결합한 항체를 통해 선택적으로 전달됩니다.
Trastuzumab emtansine(Kadcyla)은 인간화 단클론 항체 trastuzumab과 세포독성 물질 DM1이 결합된 ADC입니다. HER2 수용체에 결합하여 세포 내로 유입된 후, 리소좀에서 분해되어 DM1이 방출되며, 튜불린에 결합해 세포 분열을 억제하고 세포사를 유도합니다. HER2의 과발현 세포에 선택적으로 작용하여 표적 치료 효과를 발휘합니다.
