Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed cell surface transmembrane glycoprotein that interacts with multiple integrins and regulates their functions. Engagement of CD47 by soluble ligands or counter receptors modulates various signaling pathways, including activation of heterotrimeric G proteins. Functionally, CD47 is involved in increasing intracellular calcium concentration upon cell adhesion to the extracellular matrix.

It also serves as a receptor for the C-terminal cell-binding domain of thrombospondin and may play roles in membrane transport and signal transduction. CD47 is broadly distributed across tissues and has reduced expression on Rh erythrocytes. As a ligand for CD172a (SIRPα), an immune inhibitory receptor on macrophages, CD47 prevents phagocytosis of CD47-positive cells. The CD47–CD172a system influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, particularly in synapse-rich regions of the brain and retina.
