The Ambion™ TaqMan™ MicroRNA Cells-to-CT™ Kit enables microRNA (miRNA) expression profiling directly from cultured cells without RNA purification. It provides a simple and complete workflow for miRNA expression analysis, suitable for both small-scale and automated, high-throughput applications.

Complete, Validated Solution

This kit features a unique method for lysing cultured cells while removing genomic DNA and preserving RNA integrity. It includes TaqMan™ MicroRNA reverse transcription reagents and TaqMan™ Universal PCR Master Mix for real-time PCR analysis. The kit has been validated with TaqMan™ MicroRNA Assays (sold separately).

Simple, Convenient 10-Minute Sample Preparation

The procedure allows preparation from 10–100,000 cultured cells per sample in plates or tubes. Cells are washed in PBS, lysed at room temperature for 8 minutes, and treated with DNase concurrently. Lysis is terminated with a Stop Solution after 2 minutes. Lysates can be used immediately for RT or stored at -20°C for up to 5 months. No heating, washing, or centrifugation is required.

Gold Standard Specificity

TaqMan™ MicroRNA Assays use unique stem-loop RT primers designed for mature miRNA targets, ensuring high specificity and the ability to distinguish single-base differences. The kit’s RT reagents are optimized for this assay design, providing results equivalent to purified RNA.

Robust Performance

Validated across >900 TaqMan™ MicroRNA Assays, the kit delivers performance comparable to purified RNA for miRNA expression analysis.

Broad Input Dynamic Range

Accurately quantitate miRNA from as few as 10 cells up to 100,000 cells. Up to 33% of the RT reaction volume can consist of sample lysate, maintaining linear detection across 5 logs of input.
