Target Information

Connexin 43 (Cx43) is a member of the gap junction protein family. Connexins assemble as hexamers and form hemichannels that connect adjacent cells to create cell-to-cell channels. These clusters form gap junctions, which are crucial for intercellular communication, development, and cell growth regulation.

Phosphorylation of Cx43 by protein kinase C (PKC) at Ser368 regulates gap junction assembly and function, reducing cell-to-cell communication. Src kinase also interacts with and phosphorylates Cx43, further modulating junctional communication.

Connexin 43 is the predominant gap junction protein in cardiac tissue, essential for synchronized contraction and embryonic development. It is regulated by multiple protein kinases affecting myocardial cell-cell coupling. A related pseudogene, GJA1P, is mapped to chromosome 5. Mutations in related gap junction genes can lead to peripheral neuropathies and heart malformations. Alternatively spliced transcript variants of GFAP have also been identified.
