Product Specific Information

The 2B8 monoclonal antibody reacts with mouse CD117 (c-Kit receptor, Steel factor receptor, stem cell factor receptor), a member of the tyrosine kinase receptor family. This 145-kDa molecule is expressed by most hematopoietic progenitor cells in mouse bone marrow and also by mast cells.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse bone marrow cells; optimal use ≤ 1 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
Excitation/Emission: 488–561 nm / 775 nm; compatible with Blue, Green, Yellow-Green lasers.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

KIT (c-KIT) is a proto-oncogene and a type 3 transmembrane receptor for mast cell growth factor (MGF, stem cell factor). It regulates growth and activation of various hematopoietic and non-hematopoietic cells.
Mutations in KIT are linked to gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism.
Activation of c-Kit triggers autophosphorylation and downstream signaling (Ras/ERK, PI3K, PLC-γ, JAK/STAT), essential for hematopoiesis, stem cell maintenance, and gametogenesis.
