Product Specific Information
The 6H6 monoclonal antibody reacts with human CD123, the alpha chain of the IL-3 receptor (60–70 kDa transmembrane protein). CD123 binds IL-3 with low affinity alone, and with CD131 forms a high-affinity IL-3 receptor complex. It is expressed by myeloid precursors, macrophages, dendritic cells, mast cells, basophils, and megakaryocytes.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (1 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.
Light Sensitivity: Protect from light; tandem dye is photo-sensitive.
Fixation: Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) with minimal brightness impact.
Excitation/Emission: 488–561 nm / 775 nm; compatible with Blue, Green, and Yellow-Green lasers.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
IL-3Ra (Interleukin-3 receptor subunit alpha)
Belongs to the type I cytokine receptor family. Forms a heterodimer with the common beta chain (CDw131). Responsible for IL-3 signal transduction. IL-3Ra/IL-3Rb heterodimerization is induced by IL-3 and required for receptor activation. Overexpressed in acute myeloid leukemia (AML) blasts compared to normal hematopoietic cells, making it a potential therapeutic target.
