Product Specific Information
The monoclonal antibody B6H12 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein with a small, alternatively spliced cytoplasmic domain. It is involved in adhesion through interactions with SIRP (signal regulator protein) and is associated with β3 integrins (CD51/CD61 and CD41/CD61).
This interaction mediates bi-directional signaling to modify neural synaptic activity and regulate macrophage phagocytosis. CD47 acts as a receptor for thrombospondin, and its expression on T cells can mediate activation or apoptosis depending on thrombospondin levels.
Stimulation of CD47 by monoclonal antibody has been shown to induce CD4+CD25- suppressive activity and increase Foxp3 expression.
Expression is found in most hematopoietic cells (T, B cells, monocytes, platelets, erythrocytes) and also in non-hematopoietic cells.
This antibody has reported neutralizing activity.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells.
Use at 5 µL (0.25 µg) per test in a final volume of 100 µL. Cell number may vary from 10⁵–10⁸ cells/test.
Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information
CD47 (Integrin-associated protein, IAP) is a ubiquitously expressed cell surface transmembrane glycoprotein that interacts with several integrins and regulates their functions.
Engagement of CD47 modulates various signaling pathways, including activation of heterotrimeric G proteins and intracellular calcium increase upon cell adhesion.
CD47 is also a receptor for the C-terminal cell binding domain of thrombospondin and may play roles in membrane transport and signal transduction.
It is widely distributed across tissues, reduced on Rh erythrocytes, and acts as a ligand for CD172a (SIRPα), an inhibitory receptor on macrophages that prevents phagocytosis of CD47-positive cells.
The CD47–CD172a system influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, being abundant in synapse-rich brain and retina regions.
