Product Specific Information
This CMSSB monoclonal antibody reacts with human CD56 (Neural Cell Adhesion Molecule, NCAM), a highly glycosylated transmembrane molecule expressed by neurons and involved in homotypic neural cell adhesion.
In the hematopoietic system, CD56 is expressed on NK cells and NKT cells. Staining with CMSSB does not block binding of MEM188 or CB56.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated on normal human peripheral blood cells. Recommended 5 µL (0.06 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.
Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation; protect vial and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
Excitation: 488–561 nm; Emission: 667 nm; Laser: Blue, Green, Yellow-Green
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD56 (NCAM) is a transmembrane glycoprotein of the immunoglobulin family functioning as an adhesive molecule. It is expressed throughout the nervous system (120–180 kDa isoforms) and on T and NK cells. CD56 participates in cell migration, axonal growth, and synaptic plasticity.
Polysialic modification reduces adhesion. Through its extracellular region, CD56 mediates homophilic and heterophilic interactions with extracellular matrix components such as laminin and integrins.
CD56 is expressed on most neuroectodermal-derived cell lines, tissues, and tumors (retinoblastoma, medulloblastoma, astrocytoma, neuroblastoma), and serves as a sensitive neuroendocrine marker for neuroendocrine and ovarian granulosa cell tumors.
Diseases associated with CD56 dysfunction include rabies and blastic plasmacytoid dendritic cell neoplasms.
