Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M cell cycle arrest. Structurally, p53 contains:

• N-terminal transactivation domain
• Central DNA-binding domain
• Oligomerisation domain
• C-terminal regulatory domain

Phosphorylation at Ser15 is critical for p53 activation and stabilization. p53 acts as a tumor suppressor by preventing proliferation of damaged cells, and its dysregulation is associated with tumor formation, aging, and oxidative stress. Mutations in the TP53 gene are common in many human cancers and can result in loss of tumor suppressor function. Germline mutations in TP53 are also linked to hereditary cancer syndromes such as Li-Fraumeni syndrome.
