Thermo Scientific™ aminoxyTMTsixplex™ Label Reagent Set
The aminoxyTMTsixplex™ Label Reagents share an identical structure but contain different numbers of ¹³C and ¹⁵N isotopes in the mass reporter. The optimized labeling procedure can be completed in one hour. After tag labeling, samples are quenched and cleaned using HILIC solid-phase extraction before mass spectrometry analysis.
The six compounds of the aminoxyTMTsixplex™ Reagent Set have the same mass (isobaric) and chemical structure (carbonyl-reactive aminoxy group, spacer arm, and mass reporter). The specific distribution of ¹³C and ¹⁵N isotopes results in unique reporter masses (126–131 Da) in the low-mass region, enabling relative quantitation of labeled molecules in multiplexed MS samples representing six different conditions.
For glycobiology MS applications, these reagents enable quantitative profiling of glycan isoforms and biomarker discovery. They improve ionization and retention of labeled glycans by reversed-phase liquid chromatography (RPLC). The aminoxyTMT reagents can quantify carbohydrates, steroids, or oxidized proteins.

Native glycans are difficult to study by mass spectrometry due to poor ionization efficiency. The aminoxy group shows better reactivity with carbonyls and improved stability compared to hydrazides. Labeling with aminoxyTMT reagents enhances ionization and sensitivity, enabling reliable quantitation of glycans across multiple samples.
