Target Information
FAM175B is a component of the BRISC complex, a multiprotein complex that specifically cleaves Lys-63-linked polyubiquitin, leaving the last ubiquitin chain attached to its substrates. It may act as a central scaffold protein assembling BRISC components and retaining them in the cytoplasm.
Functions include:
- Regulation of apoptosis via modulation of Lys-63-linked ubiquitination
- Mitotic spindle assembly and microtubule attachment to kinetochores via deubiquitination of NUMA1
- Deubiquitination of interferon receptor IFNAR1, enhancing its stability and surface expression
- Down-regulation of LPS response via IFNAR1 deubiquitination
- Induction of p53/TP53 in response to DNA damage
- Promotion of USP7-p53 interaction, preventing p53 degradation and enhancing transcriptional regulation and apoptosis
