Target Information

Programmed death-ligand 2 (PD-L2, also known as B7-DC) is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it serves as a ligand for PD-1, an inhibitory immune checkpoint receptor.
PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Its expression is upregulated by cytokines such as IL-4, TNF-α, and IFN-γ.
Although expressed at lower levels than PD-L1, PD-L2 exhibits a 2–6 times higher affinity for PD-1. Binding of PD-L2 or PD-L1 to PD-1 inhibits T cell activation, proliferation, and cytokine secretion, contributing to immune regulation.
Additionally, PD-L2 can interact with RGMb (repulsive guidance molecule b), influencing respiratory tolerance and immune signaling pathways.
