Target Information

CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also mediates HIV-2 binding and fusion. CXCR4 serves as a receptor for the chemokine CXCL12/SDF-1, regulating intracellular calcium levels and MAPK1/MAPK3 activation. Additionally, it acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP signaling.
CXCR4 plays critical roles in hematopoiesis, cardiac ventricular septum formation, and gastrointestinal vascularization. In the CNS, it is involved in hippocampal neuron survival and cerebellar development. CXCR4 also binds bacterial LPS and mediates LPS-induced inflammatory responses, including TNF secretion by monocytes.
Antibodies to CXCR4 can block HIV-1 and HIV-2 fusion and infection in human target cells. The amino-terminal domain and second extracellular loop serve as HIV binding sites.
