NMD3의 돌연변이는 mRNA 분해를 담당하는 주요 세포질 엑소리보뉴클레이스인 XRN1이 결핍된 경우 치명적입니다.
NMD3 단백질 서열에는 알려진 기능적 모티프가 존재하지 않지만, NMD3 상동체들은 Cx2C 반복을 포함하는 아미노 말단 도메인을 가지고 있어, 핵산 결합 또는 단백질 올리고머화에 관여하는 것으로 추정됩니다.
