Target Information

The C4B gene encodes the basic form of complement factor 4, a key component of the classical complement activation pathway. The protein is synthesized as a single-chain precursor that is cleaved into a trimer of alpha, beta, and gamma chains prior to secretion. This trimer facilitates interactions between antigen-antibody complexes and other complement components.

The alpha chain can be further cleaved to release C4 anaphylatoxin, which mediates local inflammation. Deficiency in this protein is associated with systemic lupus erythematosus (SLE). The gene is located in the MHC class III region on chromosome 6, and exists in multiple haplotypes, allowing 1–3 gene copies per individual. Variants include long and short forms due to insertion or absence of a 6.4 kb HERV-K retrovirus in intron 9.
