Target Information
BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death. The BCL-2 family members modulate apoptosis through dimerization. For example, Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Bax and Bak are critical for cytochrome c release from mitochondria, initiating apoptosis. Bax exerts a pro-apoptotic effect, while constitutive expression of BCL-2, such as translocation to the Ig heavy chain locus, is associated with follicular lymphoma. High BCL-2 expression is observed in neoplastic germinal centers of follicular lymphomas, but not in normal or hyperplastic ones. Two transcript variants of BCL-2 differ in their C-terminal ends, and overexpression has been linked to cancers such as B-cell lymphoma and prostate cancer.
