Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change allowing dimerization and binding to estrogen response elements (ERE) in DNA. When bound to EREs, ER can positively or negatively regulate gene transcription through recruitment of coactivator or corepressor proteins.

There are two forms of estrogen receptors, alpha (ESR1) and beta (ESR2), encoded by separate genes. Alternative RNA splicing produces at least four estrogen receptor-alpha isoforms (66 kDa, 53 kDa, 47 kDa, and 35 kDa).

Estrogen receptors are widely expressed in various tissues and are essential for sexual development and reproductive function. They also play roles in bone and other tissues, and are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
