Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. Through this mechanism, ER regulates gene transcription via coactivator or corepressor proteins.
There are two forms of the estrogen receptor, alpha (ESR1) and beta (ESR2), encoded by separate genes. Multiple isoforms of estrogen receptor-alpha exist due to alternative RNA splicing (Isoforms 1–4, ranging from 35–66 kDa). ERs are widely expressed in various tissues, playing critical roles in sexual development, reproductive function, and bone health. They are also implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
