비인산화 상태에서는 비활성이나, Thr-Gly-Tyr 모티프의 이중 인산화에 의해 활성화됩니다.
p38 MAPK는 4가지 isoform을 가지며, 조직별 발현 및 상위/하위 조절인자에 대한 친화도가 다릅니다.
TNF, IL-1, 열충격 등의 자극에 의해 MAPK kinase-3/6가 Thr180과 Tyr182를 인산화시켜 p38 MAPK를 활성화합니다.
활성화된 phospho-p38 MAPK는 ATF-2, CHOP-1, MEF-2 등의 전사인자를 인산화하여 다양한 세포 반응을 유도합니다.
