Product Specific Information

Description:
The M1/70 monoclonal antibody reacts with mouse CD11b (integrin alphaM, 165–170 kDa). CD11b associates with CD18 to form the αMβ2 integrin (Mac-1), binding to CD54 (ICAM-1), C3bi, and fibrinogen. It is expressed on macrophages, NK cells, granulocytes, activated lymphocytes, and B-1 cells in the peritoneal cavity. The antibody also cross-reacts with human CD11b, detecting it on monocytes, granulocytes, and some NK cells.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes and bone marrow cells. Use ≤0.125 µg per test (defined as the antibody amount staining a 100 µL cell sample). Optimal titration is recommended.

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation/Emission:
488–561 nm / 667 nm
Laser: Blue, Green, Yellow-Green

Filtration:
0.2 µm post-manufacturing filtered.


Target Information

CD11b (integrin alpha-M, ITGAM):
A 165 kDa adhesion molecule forming a heterodimer with CD18 (integrin beta-2), known as Mac-1 or CR3. Expressed on monocytes/macrophages, granulocytes, NK cells, dendritic cells, and microglia.
Functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating cell-cell and cell-matrix interactions.
Expression varies across cell types — 8% of spleen cells, 44% of bone marrow cells, <1% of thymocytes — and is a common microglial marker.
Associated with systemic lupus erythematosus and ITGAM-related autoimmune susceptibility.
