Target Information

CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides. It is expressed on lymphoblastic, Burkitt’s, and follicular germinal center lymphoma cells, immature B cells in adult bone marrow, and cells from patients with chronic myelocytic leukemia (CML). CD10 is also found on breast myoepithelial cells, bile canaliculi, fibroblasts, and is highly expressed on the brush border of kidney and gut epithelial cells.

Functionally, CD10 cleaves peptides at the amino side of hydrophobic residues and inactivates peptide hormones such as glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. It is a 100 kDa type II transmembrane glycoprotein encoded by a single-copy gene of over 45 kb. The 5′ untranslated region of the CD10 gene is alternatively spliced, producing four distinct mRNA transcripts, though the coding region remains unaffected. Diseases associated with CD10 dysfunction include spinocerebellar ataxia 43 and Charcot-Marie-Tooth disease.
