Target Information

F-Box and WD Repeat Domain Containing 7 (FBXW7) belongs to the F-box protein family characterized by a ~40 amino acid F-box motif.
F-box proteins form part of the SCF (SKP1-cullin-F-box) ubiquitin ligase complex, which mediates phosphorylation-dependent ubiquitination.
They are grouped into three classes:

1. Fbws – containing WD-40 domains
2. Fbls – containing leucine-rich repeats
3. Fbxs – containing other interaction modules or no recognizable motifs

FBXW7 (previously FBX30) is a member of the Fbws class and contains seven tandem WD40 repeats.
It binds directly to cyclin E and likely targets it for ubiquitin-mediated degradation.
Mutations in FBXW7 are associated with ovarian and breast cancer, suggesting a role in tumorigenesis.
Multiple transcript variants encoding different isoforms have been identified.
