Target Information
The PPP2R2B gene encodes a beta isoform of the regulatory subunit B55 of protein phosphatase 2 (PP2A), one of the major Ser/Thr phosphatases involved in the negative regulation of cell growth and division. PP2A consists of a catalytic subunit and a constant regulatory subunit that associates with various regulatory subunits. The B regulatory subunit modulates substrate selectivity and catalytic activity.
Defects in PPP2R2B cause autosomal dominant spinocerebellar ataxia 12 (SCA12), a neurodegenerative disease affecting the cerebellum, sometimes involving the brainstem and spinal cord, leading to impaired coordination of speech and body movements. Multiple alternatively spliced variants exist, with some containing a CAG trinucleotide repeat sequence in the 5′ UTR that can expand in SCA12 cases.
