Target Information
Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are transported within the cell by approximately 300 kDa cation-independent mannose 6-phosphate receptors (CI-MPR). These receptors participate in both biosynthetic and endocytic cycles, moving lysosomal enzymes between the trans-Golgi network, endosomes, and the plasma membrane. MPRs cycle through these compartments roughly every 3 hours, with most located in late endosomes, fewer in the trans-Golgi network, and about 10% at the cell surface.
The MPR also binds type II insulin-like growth factor (IGF-II), allowing simultaneous binding of man6P and IGF-II. A soluble ~240 kDa truncated form representing the extracellular domain circulates in serum and retains ligand-binding capability.
