Target Information
Upon entry into mitosis, the cell’s microtubule (MT) network forms the mitotic spindle, enabling chromosome segregation. Kinetochores (KT) on centromeric chromatin are essential for proper attachment of chromosomes to spindle MTs. The spindle and kinetochore complex (SKA), composed of SKA1, SKA2, and SKA3, is required for stable KT-MT interactions and timely anaphase onset.
Depletion of SKA3 by siRNA delays anaphase transition, resulting in a prolonged metaphase-like state. SKA3-depleted cells accumulate high levels of checkpoint protein Bub1 at kinetochores, indicating that the SKA complex is crucial for spindle checkpoint silencing and chromosome cohesion maintenance during mitosis.
