- Immunohistochemistry (PFA fixed):
Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Ep-CAM (epithelial adhesion molecule, epithelial specific antigen, ESA) is a transmembrane glycoprotein expressed in epithelial tissues (~40 kDa). It functions as a Ca²⁺-independent homotypic cell adhesion molecule, influencing cell cycle, proliferation, and metabolism via induction of proto-oncogene c-myc and cyclins A/E.
Ep-CAM-mediated adhesions negatively regulate cadherin-mediated adhesions, affecting epithelial differentiation and growth. Overexpression is linked to enhanced epithelial proliferation and is highly expressed in human carcinomas, serving as a marker for epithelial lineage tumors.
Ep-CAM is found on the basolateral surface of most simple epithelia and many carcinoma types, and can distinguish adenocarcinomas from pleural mesotheliomas.
