GOLPH3는 처음에 trans-Golgi network에 위치한 주변막 단백질로 확인되었으나, 이후 미토콘드리아 특이적 인지질인 cardiolipin을 조절하여 미토콘드리아 질량을 조절하는 단백질로 보고되었습니다.
GOLPH3는 TOR 신호전달 경로에 관여하며, 과발현 시 세포 증식 및 anchorage-independent growth를 증가시킵니다. 또한 EGF 자극에 의해 S6 kinase 활성과 AKT 인산화를 증가시키며, siRNA 처리 시 이러한 신호가 억제됩니다.
이러한 결과는 GOLPH3가 bona fide oncogene으로 작용하며, 치료 전략의 유용한 타겟이 될 수 있음을 시사합니다.
