TSEN2는 tRNA-splicing endonuclease 복합체의 두 촉매 소단위 중 하나로, pre-tRNA의 스플라이스 부위를 인식하고 절단하는 역할을 합니다. 이 효소는 pre-tRNA를 절단하여 인트론과 두 개의 tRNA 절반 분자를 생성하며, 각각 2’,3’-cyclic phosphate 및 5’-OH 말단을 가집니다. 스플라이스 부위에는 보존된 서열이 없지만, 인트론은 항상 동일한 위치에 존재하여 일정한 구조적 간격을 유지합니다.
Isoform 1은 5’-스플라이스 부위 절단의 활성 부위를 포함하며, Isoform 2는 다른 RNA 기질을 처리하지만 pre-tRNA 절단 능력은 없습니다. 이 복합체는 pre-mRNA 3’-말단 가공 인자와 연관되어 tRNA 및 mRNA 처리 과정 간의 연계를 형성합니다.
