Target Information
ICAD (Inhibitor of Caspase-Activated DNase) is a substrate that regulates nuclear apoptosis. It exists in two isoforms: a functional 45 kDa form (ICAD-L/DFF45) and a 35 kDa form (ICAD-S/DFF35). While both isoforms can bind and inhibit CAD, only ICAD-L is functional. During apoptosis, ICAD is cleaved by caspase-3, allowing CAD to fragment DNA. In non-apoptotic cells, CAD remains complexed with ICAD. ICAD also serves as a folding chaperone for CAD. Recent studies suggest ICAD as a potential therapeutic target in colon and brain cancers. Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
