Target Information
Plasma prekallikrein is a glycoprotein involved in surface-dependent activation of blood coagulation, fibrinolysis, kinin generation, and inflammation. It is synthesized in the liver and secreted into the blood as a single polypeptide chain. Upon activation by factor XIIa, plasma prekallikrein is converted to plasma kallikrein by cleavage of an internal Arg-Ile bond.
Plasma kallikrein consists of a heavy chain and a light chain linked by a disulfide bond. The heavy chain contains four tandem repeats of approximately 90 amino acids, each forming an apple domain, which contributes to its pro-coagulant activity. The light chain carries the catalytic domain homologous to the trypsin family of serine proteases. Deficiency of plasma prekallikrein can cause prolonged activated partial thromboplastin time in patients.
