Target Information
TNIK (TRAF2 and NCK interacting kinase) is characterized by an N-terminal kinase domain and a C-terminal GCK domain that serves a regulatory function. TNIK is mainly expressed in brain, heart, and spleen and acts as a specific effector of RAP2, regulating the actin cytoskeleton. TNIK undergoes autophosphorylation dependent on lys54 in the ATP-binding pocket of its kinase domain and plays a key role in cytoskeleton regulation. It also phosphorylates gelsolin, the principal actin-severing protein, suggesting TNIK’s function in cytoskeletal organization.
