SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2), also known as 2019-nCoV, is a virus causing illnesses from mild cold symptoms to severe respiratory distress. The Spike (S) protein is crucial for viral attachment, fusion, and entry, serving as a key target for antibody, inhibitor, and vaccine development.
The receptor-binding domain (RBD) within the S1 subunit binds strongly to human and bat ACE2 receptors, exhibiting higher affinity compared to SARS-CoV.
SARS-CoV-2 S protein (1273 amino acids) includes:
- Signal peptide (1–13 aa)
- S1 subunit (14–685 aa) – receptor binding
- S2 subunit (686–1273 aa) – membrane fusion
The S-ECD represents the extracellular domain of the Spike protein.
