Target Information
ICAD (Inhibitor of Caspase-Activated DNase) is a substrate that regulates nuclear apoptosis by inhibiting CAD (Caspase-Activated DNase).
ICAD has two isoforms:
- ICAD-L/DFF45 (45 kDa): Functional isoform
- ICAD-S/DFF35 (35 kDa): Non-functional isoform
ICAD-L binds and inhibits CAD, but is cleaved by caspase-3 at residues 117 and 224 during apoptosis, releasing CAD to execute DNA fragmentation.
In non-apoptotic cells, CAD remains complexed with ICAD.
ICAD also functions as a CAD-specific folding chaperone.
Recent studies suggest ICAD may be a potential therapeutic target for restoring normal apoptotic signaling in colon and brain cancer cells.
Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
