Target Information
Beta Arrestin 1은 중추신경계에서 특히 풍부하게 발현되는 단백질군의 구성원으로, 어댑터 또는 스캐폴드 분자로 작용합니다. 이 단백질은 mitogenic signaling에 필수적이며, G protein-coupled receptors (GPCRs, 예: β2-adrenergic receptor)의 작용제 의존적 탈감작 및 내재화를 매개합니다. GPCR은 리간드 결합 후 G-protein receptor kinases (GRKs)에 의해 세린 잔기에 인산화되고, 세포질 내 beta Arrestin 1은 ERK1/2에 의해 Ser412에서 인산화되어 음성 피드백 메커니즘을 통해 인산화된 수용체에 결합합니다. 이후 Ser412가 탈인산화되고 GPCR이 내재화되어 Ras, Raf, ERK1/2 신호경로가 활성화됩니다.
