Target Information
Cas9은 CRISPR(Clustered Regularly Interspaced Short Palindromic Repeats) 시스템과 연관된 RNA-유도 DNA 엔도뉴클레이스 효소로, 박테리아의 적응 면역 시스템 및 유전자 공학 연구에 사용됩니다.
RNA를 통해 원하는 DNA 서열을 표적화할 수 있어 유연성이 높으며, indel 변이 유도, 특정 서열 삽입/치환, 대규모 유전체 재배열 등 다양한 유전체 편집에 활용됩니다.
Cas9은 Streptococcus pyogenes (SpCas9) 또는 Staphylococcus aureus (SaCas9)에서 유래할 수 있습니다. SaCas9은 작은 크기, 높은 효율, nickase 활성, 높은 특이성 등의 장점을 가지며, NNGRRT PAM을 인식하고 SpCas9과 유사한 수준의 DNA 절단 효율을 보입니다.
SaCas9은 SpCas9(1368 AA)보다 작고, NGGRRT PAM을 가진 표적 DNA에서 유사한 indel 생성률을 보여 AAV 기반 올인원 전달 시스템에 적합합니다.
