Target Information
EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to environmental stresses. It plays a key role in antiviral defense through activation by double-stranded RNA produced during viral infection. PKR is involved in IFN-induced antiviral response, virus-induced apoptosis, and cell growth regulation. It mediates apoptosis via Fas and FADD/Caspase-8 signaling and regulates pathways including NF-κB, p53, p38, and PDGF. PKR has been linked to tumor suppression, malignancy, and neurodegenerative diseases such as Alzheimer’s, Huntington’s, and ALS. Viruses can inhibit PKR activity, e.g., influenza virus NS1 protein.
