Raf-1 (c-Raf)은 Ras 계열의 막 결합 GTPase 하류에서 작용하는 MAP kinase kinase kinase (MAP3K)입니다. 활성화된 Raf-1은 MEK1 및 MEK2를 인산화하여 활성화시키며, 이들은 다시 ERK1 및 ERK2를 인산화하여 세포 생리 조절에 관여합니다. 활성화된 ERK는 유전자 발현, 세포 분열, 세포사멸(apoptosis), 분화, 이동 등에 중요한 역할을 합니다.
Raf-1 유전자 변이는 Noonan syndrome 5 및 LEOPARD syndrome 2와 관련되어 있습니다.
