This gene belongs to the Abd-B homeobox family and encodes a protein with a homeobox DNA-binding domain. It is part of the homeobox D gene cluster on chromosome 2. The encoded nuclear protein acts as a sequence-specific transcription factor expressed in developing limb buds, contributing to differentiation and limb development. Mutations in this gene are associated with Wilms' tumor and congenital vertical talus, as well as foot deformities resembling those in Charcot-Marie-Tooth disease.
