Target Information
NPM1 (Nucleophosmin 1, B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in ribosome assembly and transport, cytoplasmic/nuclear trafficking, regulation of DNA polymerase alpha activity, centrosome duplication, and regulation of p53.
It shuttles between the nucleus, cytoplasm, and nucleolus, acting as a chaperone for core histones. NPM1 binds nucleic acids, prevents protein aggregation, protects enzymes during thermal stress, and assists in renaturation of denatured proteins.
NPM1 is associated with centrosome duplication and serves as a substrate for CDK2/cyclin E. It also forms part of several fusion proteins (NPM-ALK, NPM-RAR, NPM-MLF1) linked to tumorigenesis and sequesters tumor suppressor RF in the nucleolus.
Dysfunction of NPM1 is associated with diseases such as acute myeloid leukemia and lymphomatoid papulosis.
