Description:
The TRFK5 antibody reacts with mouse and human interleukin-5 (IL-5) and functions as a neutralizing antibody. Mouse IL-5 is a disulfide-linked homodimer composed of two 113 amino acid peptides (32–34 kDa in SDS-PAGE). IL-5 is produced by T cells and is also known as eosinophil-differentiating factor (EDF), B cell growth factor II (BCGFII), and T cell-replacing factor (TRF). It induces eosinophil differentiation, survival, and activation, and in mice, it stimulates B cell proliferation and antibody production.

Applications Tested:
The Functional Grade Purified TRFK5 antibody has been tested by LAL assay for low endotoxin levels, by ELISA, and in bioassay for neutralization of mouse IL-5 bioactivity.
At 0.06 µg/mL, the antibody inhibits 50% of the biological effects of 1.0 ng/mL mouse IL-5 in an MC/9 cell proliferation assay.

ELISA Usage:

• Mouse IL-5: Capture antibody (1–4 µg/mL) with biotin TRFK4 (13-7051) for detection and recombinant mouse IL-5 (14-8051) as standard.
• Human IL-5: Capture antibody (1–4 µg/mL) with biotinylated JES1-5A10 (13-7059) for detection and recombinant human IL-5 (14-80591) as standard.
• Include a standard curve with doubling dilutions from 1000 pg/mL to 8 pg/mL.

Storage and Handling:

• Use in a sterile environment.
• 0.2 µm post-manufacturing filtered.
• Purity: >90% (SDS-PAGE).
• Endotoxin Level: <0.001 ng/µg antibody (LAL assay).
• Aggregation: <10% (HPLC).

Target Information

IL-5 is a cytokine that acts as a growth and differentiation factor for B cells and eosinophils. It is a major regulator of eosinopoiesis, eosinophil maturation, and activation. Elevated IL-5 levels are associated with asthma and hypereosinophilic syndromes.
The IL-5 receptor is a heterodimer whose beta subunit is shared with receptors for IL-3 and GM-CSF (CSF2). IL-5, IL-4, IL-13, and CSF2 form a cytokine gene cluster on chromosome 5q31, regulated by long-range regulatory elements.
