• For IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.

Target Information

The F13A1 gene encodes the coagulation factor XIII A subunit, the final zymogen activated in the blood coagulation cascade. Plasma factor XIII is a heterotetramer of two A subunits (catalytic) and two B subunits (non-enzymatic carriers). Platelet factor XIII consists of two A subunits identical to those in plasma. Upon thrombin cleavage and in the presence of calcium ions, the B subunits dissociate, yielding active factor XIIIa. This enzyme functions as a transglutaminase, catalyzing γ-glutamyl-ε-lysine crosslinks between fibrin molecules, stabilizing the fibrin clot. It also crosslinks α2-plasmin inhibitor or fibronectin to fibrin α chains.
Deficiency of factor XIII can be:

• Type I: Lack of both A and B subunits
• Type II: Lack of A subunit only
  Such deficiencies result in prolonged bleeding, impaired wound healing, and recurrent miscarriage.
