Target Information
STK3 and STK4 are stress-activated, pro-apoptotic kinases that, following caspase cleavage, enter the nucleus and induce chromatin condensation and DNA fragmentation. They are key components of the Hippo signaling pathway, which controls organ size and tumor suppression by restricting proliferation and promoting apoptosis.
The core of this pathway is a kinase cascade in which STK3/MST2 and STK4/MST1, in complex with SAV1, phosphorylate and activate LATS1/2 (in complex with MOB1). LATS1/2 then phosphorylate and inactivate YAP1 and WWTR1/TAZ, preventing their nuclear translocation and regulating genes involved in proliferation, cell death, and migration.
STK3/MST2 and STK4/MST1 repress hepatocyte proliferation, prevent activation of adult liver stem cells, and inhibit tumor formation. They also phosphorylate NKX2-1 and NEK2, regulating centrosome disjunction and localization. In conjunction with SAV1, they activate ESR1 transcriptional activity through phosphorylation modulation, positively regulate RAF1 activation, and phosphorylate MOBKL1A, RASSF2, and MOBKL1B to activate STK38.
