Target Information

Proposed scaffold protein implicated in various physiological processes, partly through regulation of ubiquitination of specific cellular proteins. It modulates cullin-RING E3 ubiquitin ligase (CRL) complexes by displacing CAND1, promoting CRL E3 activity and dissociating CAND1 from CUL1 and CUL2. COMMD1 promotes ubiquitination and proteasomal degradation of NF-κB subunit RELA, leading to down-regulation of NF-κB activity.

It also regulates membrane expression and ubiquitination of SLC12A2, modulates Na⁺ transport via ENaC subunit ubiquitination, and promotes CFTR surface expression. Additionally, COMMD1 influences copper ion homeostasis and ATP7A trafficking, likely through its role in the CCC complex and cooperation with the WASH complex on early endosomes. It can bind one copper ion per monomer and may facilitate biliary copper excretion within hepatocytes. Furthermore, it interacts with phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) and regulates HIF1A-mediated transcription by competing with ARNT/Hif-1-beta.
