Target Information
PSME4 is an associated component of the proteasome that specifically recognizes acetylated histones and promotes ATP- and ubiquitin-independent degradation of core histones during spermatogenesis and the DNA damage response.
It binds acetylated histones via its bromodomain-like (BRDL) region and activates the proteasome by opening the gated channel for substrate entry.
PSME4 interacts with the core proteasome via its C-terminus, occupying the same binding sites as proteasomal ATPases and promoting an active gating mechanism.
It is a component of the spermatoproteasome, a testis-specific form of the proteasome that facilitates histone exchange during spermatogenesis, and also functions in somatic cells during DNA damage response by promoting histone degradation following double-strand breaks.
