Target Information
CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, and also participate in ubiquitin-mediated receptor removal from the cell surface.
CD33 is expressed on myelomonocytic lineage cells and binds sialic acid residues on cell surface glycans. It is a therapeutic target for acute myeloid leukemia and is found on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells. CD33 serves as a useful marker for peripheral monocytes and for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
