Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, and participates in receptor removal via ubiquitin-mediated processes.
CD33 is expressed on myelomonocytic lineage cells, granulocyte and macrophage precursors, and peripheral monocytes, but not on pluripotent stem cells. It serves as a marker for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias and is a therapeutic target for acute myeloid leukemia.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
