Target Information
The NF-kB/Rel transcription factors are present in the cytosol in an inactive state, complexed with inhibitory IkB proteins. Activation of NF-kB commonly occurs through phosphorylation-induced, proteasome-mediated degradation of IkB. The key regulatory step involves activation of the IkB kinase (IKK) complex, composed of IKK alpha, IKK beta (catalytic subunits), and IKK gamma (regulatory subunit). Activation of IKK occurs via phosphorylation at Ser177 and Ser181 in IKK beta (Ser176 and Ser180 in IKK alpha), leading to conformational changes and kinase activation.
