Target Information

The tumor necrosis factor superfamily member TNFSF4 (OX40L) is a type II membrane-bound, non-covalently linked homotrimeric protein. It is expressed on antigen-presenting cells such as dendritic cells and activated B-cells, as well as on vascular endothelial cells, mast cells, and natural killer cells. TNFSF4 signals through the TNFRSF4 receptor, which is expressed predominantly on CD4+ T cells and certain activated CD8+ T cells. The TNFRSF4/TNFSF4 interaction provides a costimulatory signal essential for effective communication between antigen-presenting cells and T cells, enhancing cell proliferation, survival, and cytokine expression (RANTES, IL-2, IL-3, IFN-γ). This signaling pathway plays a key role in immunotolerance.
