Target Information
SRC2 (steroid receptor coactivator 2), also known as nuclear receptor coactivator 2 (NCOA2 or TIF2), functions as a coactivator for nuclear hormone receptors. These receptors are conditional transcription factors that regulate gene expression related to cell growth, development, and homeostasis.
Members of the nuclear hormone receptor superfamily include steroid receptors and class II nuclear receptors, characterized by three domains:
- N-terminal transcriptional activation domain
- Central DNA-binding domain
- C-terminal hormone-binding domain
Before hormone binding, steroid receptors remain inactive in a complex with heat-shock proteins. Hormone binding triggers conformational changes that release the receptors, allowing them to bind DNA enhancer elements as homodimers and regulate target gene transcription. Coactivator proteins like SRC2 are required to mediate transcriptional activation.
