Product Specific Information

This CXNFT monoclonal antibody reacts with mouse NOS2 (inducible NOS, iNOS). Nitric oxide synthase enzymes catalyze nitric oxide formation from L-arginine via an NADPH- and oxygen-dependent mechanism.

• NOS Isoforms: NOS1 (nNOS), NOS2 (iNOS), NOS3 (eNOS)
• Expression: NOS2 is induced by bacterial endotoxins and inflammatory cytokines (e.g., IFN-γ, TNF-α).
• Cell Types: Expressed in M1 macrophages and myeloid-derived suppressor cells, not in M2 macrophages.
• Functional Form: Active as homodimer in presence of Ca²⁺.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on LPS-stimulated mouse peritoneal exudate cells using Intracellular Fixation and Permeabilization Buffer Set (#88-8824-00).
Recommended Usage: ≤0.06 µg per test (100 µL final volume, 10⁵–10⁸ cells/test).
Alternative Buffer Set: Foxp3/Transcription Factor Staining Buffer Set (#00-5523-00).

Fixation Compatibility:
Samples can be stored in IC Fixation Buffer (#00-8222) or 1-step Fix/Lyse Solution (#00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Filtration: 0.2 µm post-manufacturing filtered.

iNOS produces nitric oxide (NO), a gaseous free radical involved in vasorelaxation, neurotransmission, and cytotoxicity.

• Expression: Liver, macrophages, hepatocytes, synoviocytes, smooth muscle cells
• Induction: Triggered by LPS and cytokines (IFN-γ, TNF, IL-1, IL-2)
• Function: Catalyzes oxidation of L-arginine to L-citrulline and NO
• Isoforms: nNOS (Type I), iNOS (Type II), eNOS (Type III)
• Cofactors: Calmodulin, NADPH, FAD, FMN
• Clinical Relevance: Dysregulation linked to achalasia and impotence
• Genetics: Three iNOS pseudogenes located on chromosome 17 (Smith-Magenis region)
